We are pleased to share that a new research article co-authored by our colleague Carl-Magnus Svensson has been accepted for publication. The study, “Mapping the nonribosomal specificity code through promiscuity-guided A-domain engineering”, investigates how nonribosomal peptide synthetases (NRPSs) determine substrate specificity during the biosynthesis of bioactive peptides.
Using hydroxamate assays and mutant libraries of the adenylation (A-)domain in surfactin synthetase, the authors explore how enzyme promiscuity shapes substrate specificity. Their results reveal key sequence positions that strongly influence specificity and provide a roadmap for engineering NRPS systems to design new bioactive peptides.
